Gender verification in sports by PCR amplification of SRY and DYZ 1 Y chromosome specific sequences : presence of DYZ 1 repeat in female athletes
نویسنده
چکیده
Objective-To perform genetic sex typing during the Barcelona Olympic Games using polymerase chain reaction (PCR) amplification of Y chromosome specific sequences. Methods-The assay consisted of the amplification of a specific sequence corresponding to the repeat DYZI element from buccal smears samples of 2406 female competitors. Positive samples were reanalysed for the presence of another Y chromosome specific gene, SRY. Results-The expression of these two elements did not always correlate; six samples were found where the presence of DYZI but not SRY was detected. This presence ofDYZI sequence in female athletes is higher than in unselected females, where no DYZ1 amplification was observed in any of the 1629 samples analysed. Conclusions-Amplification of DYZI repeat should not be used as the only index for determining genetic sex, at least in sporting events. متن کاملAn Effective Method for Detecting Y-chromosome Specific Sequences of Circulating Fetal DNA in Maternal Plasma During the First-trimester
Background and Aims: New advances in the use of cell-free fetal DNA (cffDNA) in maternal plasma of pregnant women has provided the possibility of applying cffDNA in prenatal diagnosis as a non-invasive method. One of the applications of prenatal diagnosis is fetal gender determination. Early prenatal determination of fetal sex is required for pregnant women at risk of X-linked and some endocrin...
متن کاملGender identification of mouse embryos using the Polymerase Chain Reaction (PCR) technique
A multiplex nested PCR technique was used to identify gender from single blastomeres biopsied from 8-cell mouse embryos. The primers amplified sequences of the Sry and Zfy genes on the Y-chromosome, and a polymorphic X chromosome microsatellite locus (DXNds-3). Amplification of male DNA gave three bands, of sizes 217 bp (Zfy), 147 bp (Sry) and 111 bp (Nds). In contrast, amplification of female ...
متن کاملارزیابی آزمون تکثیر هم دمای وابسته به حلقه جهت تعیین جنسیت جنین در انسان
Background and purpose: In humans, SRY (sex-determining region of the Y chromosome) is the major gene for the sex determination which is found in normal XY males and rarely in XX males, and it is absent from normal XX females and from many XY females. There are several methods which can indicate a male genotype by amplification of SRY gene. The aim of this study was identification of the SRY ge...
متن کاملA PCR-based sex determination method for possible application in caprine gender selection by simultaneous amplification of the Sry and Aml-X genes.
Sex determination of livestock is performed to achieve the objectives of livestock breeding programmes. Techniques for sex determination have evolved from karyotyping to detecting Y-specific antigens and recently to the polymerase chain reaction (PCR), which appears to be the most sensitive, accurate, rapid and reliable method to date. In this study, a PCR-based sex determination method for pot...
